I don’t think that the Touch Screen 2 or overlays are problem here.  
Since you are using Raspberry Pi 4B - this is the [USB boot problem](https://community.volumio.com/t/guide-prepare-raspberry-pi-for-boot-from-usb-nvme/65700/46) you are facing.
